簡(jiǎn)介:中南大學(xué)博士學(xué)位論文腦膠質(zhì)瘤的表觀遺傳學(xué)研究1LRRC4在腦膠質(zhì)瘤中失活的表觀修飾的分子機(jī)制2腦膠質(zhì)瘤DNA甲基化譜的初步構(gòu)建姓名張祖萍申請(qǐng)學(xué)位級(jí)別博士專業(yè)病理學(xué)與病理生理學(xué)指導(dǎo)教師李桂源20091001博士學(xué)位論文中文摘要制,我們開展了LRRC4基因的轉(zhuǎn)錄調(diào)控機(jī)制研究。采用高特異性的啟動(dòng)子預(yù)測(cè)軟件PROMOTERLNSPECTOR和PROMOTERSCAN對(duì)其啟動(dòng)子進(jìn)行了預(yù)測(cè)分析,同時(shí)采用CPGPLOT軟件對(duì)LRRC4基因5’端調(diào)控區(qū)的CPG島進(jìn)行了分析,三者預(yù)測(cè)結(jié)果大部分重疊,綜合分析我們初步鎖定LRRC4基因啟動(dòng)子區(qū)可能位于其翻譯起始位點(diǎn)上游2151BP至101BP以內(nèi),因此我們?cè)O(shè)計(jì)引物擴(kuò)增LRRC4基因.2475A101的調(diào)控區(qū)片段,并構(gòu)建成熒光素酶報(bào)告載體PGL32475/.101。熒光素酶活性分析系統(tǒng)結(jié)果表明,PGL32475A101在COS7和HELA細(xì)胞具有與PGL3.CONTROL幾乎同等活性,這一結(jié)果表明LRRC4基因.2475/.101區(qū)域內(nèi)包含了LRRC4基因的啟動(dòng)子片段。為了進(jìn)一步定位LRRC4基因的啟動(dòng)子片段,以PGL32475/.101為模板,分別在它的5’或3’端缺失部分序列,構(gòu)建了一系列缺失突變體熒光素酶的報(bào)告載體,并將它們分別轉(zhuǎn)染COS7和HELA細(xì)胞中,發(fā)現(xiàn).835A293區(qū)域是LRRC4基因發(fā)揮啟動(dòng)子活性的必需序列,缺失這一區(qū)域LRRC4基因啟動(dòng)子則喪失了全部的啟動(dòng)活性。為了驗(yàn)證該結(jié)果我們又構(gòu)建了PGL3.835/.293/EGFP報(bào)告載體,轉(zhuǎn)染COS7和HELA細(xì)胞,發(fā)現(xiàn)LRRC4基因的.835/293區(qū)域能驅(qū)動(dòng)EGFP在COS7和HELA細(xì)胞中的表達(dá),從另一方面證明的一835/.293區(qū)域的啟動(dòng)活性。總之,通過以上研究我們成功地定位和克隆了LRRC4基因的啟動(dòng)子。LRRC4基因啟動(dòng)子在膠質(zhì)瘤中呈高甲基化狀態(tài)】生物信息學(xué)分析LRRC4基因的啟動(dòng)子序列,發(fā)現(xiàn)該序列不含有TATA盒和CAAT盒,GC含量高達(dá)70%左右,為一典型的CPG島,LI

簡(jiǎn)介:胸存大意,筆下生輝胸存大意,筆下生輝論王學(xué)仲論王學(xué)仲現(xiàn)代現(xiàn)代文人文人畫之追求畫之追求ONPURSUITOFMODERNLITERATIPAINTINGACASESTUDYOFWANGXUEZHONG專業(yè)美術(shù)學(xué)作者姓名萬(wàn)倩如指導(dǎo)教師孫國(guó)喜教授天津大學(xué)建筑學(xué)院二零一五年五月摘要摘要中國(guó)文人畫產(chǎn)生于晉代,興于宋元,至明清而大盛。最初,文人畫是被用來(lái)區(qū)別宮廷畫師和民間畫匠的作品,隨著歷史的發(fā)展,藝術(shù)家越來(lái)越注重創(chuàng)作中感情、個(gè)性成分的表達(dá)及對(duì)于畫面中氣韻與意境的追求。在二十世紀(jì)八十年代中期的西化風(fēng)潮中,部分目光長(zhǎng)遠(yuǎn)的藝術(shù)家們開始反思國(guó)畫的當(dāng)代藝術(shù)價(jià)值,一股回歸民族傳統(tǒng)的思潮被重新提出。出于對(duì)傳統(tǒng)樣式的繼承與民族情感的表達(dá),“新文人畫”的概念被提出,藝術(shù)家們?cè)噲D用現(xiàn)代的筆墨表達(dá)古人的情趣。雖以“新”為名,卻不免落入了追求形式的俗套。王學(xué)仲先生提出了“現(xiàn)代文人畫”的概念,從畫面的立意、體格、意境、哲學(xué)內(nèi)涵等方面對(duì)現(xiàn)代文人畫進(jìn)行了系統(tǒng)的歸納、概括。他將哲理蘊(yùn)涵其中,賦予現(xiàn)代文人畫新的時(shí)代面貌與詩(shī)學(xué)意境。本文通過王學(xué)仲先生的畫作及其相關(guān)研究成果,探究其作品風(fēng)格、藝術(shù)思想和藝術(shù)內(nèi)涵,并努力挖掘王學(xué)仲先生的現(xiàn)代文人畫價(jià)值及影響。本文的研究思路主要是在現(xiàn)有的理論資料基礎(chǔ)上進(jìn)行歸納與總結(jié),對(duì)王學(xué)仲先生現(xiàn)代文人畫作品中的具體畫作進(jìn)行分析研究的同時(shí),進(jìn)一步研究相關(guān)論據(jù)加強(qiáng)例證,來(lái)探求王學(xué)仲先生現(xiàn)代文人畫的藝術(shù)特點(diǎn)。同時(shí),也希望通過研究王學(xué)仲先生的現(xiàn)代文人畫作品,能對(duì)于現(xiàn)代文人畫研究的大課題有新的啟發(fā),為今后的現(xiàn)代文人畫理論研究提供借鑒。關(guān)鍵詞關(guān)鍵詞王學(xué)仲先生;現(xiàn)代文人畫;野逸美
